KEY DETAILS
Dated October 27, 2026, a mazdutide trial for atrial fibrillation after ablation is not yet recruiting, with a listed start of October 25, 2026 and completion of December 30, 2028.
An October 27, 2026 meta-analysis found CagriSema's edge over dose-matched semaglutide in type 2 diabetes clearest for body weight.
A JAMA Network Open study dated October 1, 2026 tied 2019 ADA guideline timing to moderately more guideline-concordant GLP-1 prescribing.
In Medicare patients with type 2 diabetes, an October 1, 2026 study found GLP-1 use tracked metabolic risk more than cancer status.
A citizen petition posted October 27, 2026 asks FDA not to allow compounding with seven peptide substances its advisers considered in July.
Here is where things stand. This update covers five developments. The headline item is an October 27, 2026 meta-analysis in which CagriSema's advantage over dose-matched semaglutide in type 2 diabetes was more evident for body weight. It may help to read each item closely, because the details vary a lot from one item to the next.
This one is still only a plan. Beijing Anzhen Hospital is the sponsor of the trial, registered on ClinicalTrials.gov as NCT07897305. Its listed start date is October 25, 2026, and its listed completion date is December 30, 2028. The registration describes a multicenter, open-label, superiority randomized controlled trial comparing weekly mazdutide injections with conventional routine care in adults aged 18 to 75 who have abdominal obesity and non-valvular atrial fibrillation and who regained normal heart rhythm after their first catheter ablation. The goal is to compare how well each approach prevents heart rhythm recurrence within 12 months after ablation, not counting a 3-month early recovery blanking period. Because the trial is not yet recruiting, it has no results.
The overall picture is fairly clear. The Bayesian meta-analysis, published in Diabetes, Obesity and Metabolism, found that CagriSema provides substantial glycaemic and body-weight benefits versus placebo. Against dose-matched semaglutide, the authors found its incremental advantage was more evident for body-weight reduction. Some questions remain open, though. The authors report that comparative gastrointestinal tolerability and discontinuation risk remain uncertain.
This study looks back several years. Published in JAMA Network Open on October 1, 2026, it is described by its authors as a cross-sectional study. They found that the timing of the 2019 ADA guideline update was associated with substantially increased guideline-concordant SGLT2 inhibitor prescribing and moderately increased guideline-concordant GLP-1 receptor agonist prescribing by 2020, among guideline-targeted patient subgroups compared with nontargeted ones. The SGLT2 inhibitor increases were also associated with higher first-year Longitudinal Knowledge Assessment scores measured afterward.
This one is about who receives a prescription. The retrospective study of Medicare beneficiaries with type 2 diabetes, published in JAMA Network Open on October 1, 2026, found that GLP-1 receptor agonist use increased substantially. That use was primarily associated with metabolic risk profile, with limited associations with cancer status. There is one further finding. The authors report that variation across cancer types and stages suggests oncologic context may influence prescribing.
This is a request, not a decision. The citizen petition, docket FDA-2026-P-13366, was posted on Regulations.gov on October 27, 2026. It asks the FDA to refrain from allowing the compounding of drug products using any of the seven peptide bulk drug substances considered at the FDA's Pharmacy Compounding Advisory Committee on July 23 and 24, 2026.
That covers this update. Each of the five entries links to its original listing on ClinicalTrials.gov, PubMed or Regulations.gov, where the details can be read directly.
